Habitat colonization by neophyte Impatiens glandulifera and estimate of factors limiting its spread

Invasive spread of neophyte Impatiens glandulifera in central Europe started approximatelly eighty years ago. First records of dense cover stands come from belt stands in riparian habitats. The scale of invaded habitats and degree of the dominance of I. glandulifera is more diversified nowadays. This thesis is objected on the dominance and fertility of I. glandulifera within different habitat types and scale of invaded habitats in different parts of invaded range within Europe (i. e. in Czech Republic and Switzerland). The results show that the height and cover (substitutes for biomass and dominance) of this neophyte (i) correlates with the character of invaded vegetation (ii) relates to the degree of hemeroby (a measure of human impact) negativelly, and (iii), of course, both the growth and dominance are positively affected by nutrient content. Fertility does not differ among the types of invaded habitats, but goes up with the height of I. glandulifera and decreases with its cover. Invaded habitats comprises ruderal and riparian vegetation, but also wet maedows, forest clearances, beach and slope forests or weed vegetation.
Genetic interactions of the Prdm9 gene

The Prdm9 gene (PR domain containing 9, Meisetz, Hybrid sterility 1) encodes enzyme that trimethylates histone 3 on lysines 4 and 36. These methylation marks determine the positions of DNA double-strand breaks that are repaired by meiotic homologous recombination. In this study, we assayed genetic interactions of Prdm9 with two genes important for spermatogenesis - Mili (Piwil2) involved in piRNA biogenesis and Mybl1 encoding transcription factor that regulates many genes important for prophase I, including piRNA precursors. We crossed laboratory mice carrying mutation in Prdm9 with heterozygotes for mutation in Mybl1 or Mili, and created compound heterozygotes and, in case of Mybl1, also double homozygotes. We assessed body weight and male fertility parameters (weight of testes, sperm count, malformed sperm, percentage of tubules containing spermatocytes and of abnormal nuclei of pachytene spermatocytes) of these mice and compared them to controls. We also investigated the effect of Mybl1 and Mili mutations on fecundity of F1 intersubspecific hybrids. Our results revealed possible interactions of Prdm9 and Mybl1 in the laboratory mouse. Economic aspects of screening for thyroid disease in pregnancy and in women with fertility disorders

The incidence of thyroid diseases in pregnancy in the Czech Republic reaches 10- 15%. Emphasis on early diagnosis and treatment is laid not only during pregnancy but also in the time preceding conception due to the impact of thyroid diseases on fertility, the course of pregnancy, birth and fetal development. The aim of the dissertation was to assess the effectiveness and economical aspects of screening for thyroid disease in pregnancy and in women with fertility disorders in the conditions of the Czech Republic. The dissertation consists of four published studies. The first study is a prospective cross-sectional study of 200 positively screened pregnant women. In the study we come to conclusion that pregnant women who are at high- and low-risk for thyroid disease have similar clinical and laboratory characteristics and screening, currently focused only on risk groups, is ineffective. The second study of 5 223 pregnant women is a case-control study. We find that the age of women over 30 cannot be regarded as a risk factor for thyroid disease in pregnancy, although addition this age criterion to the case-finding screening strategy improve its efficiency and ATA (American Thyroid Association) include it in their guideline 2011. Inflammatory Bowel Diseases in pregnancy

(EN) Inflammatory bowel disease is a chronic autoimmune disease of the intestinal tract including Crohn's disease, ulcerative colitis, and other unspecified colitis. Depending on the type of disease, inflammation affects the small or large intestine or rectum. It is a multifactorial disease in which the intestinal microflora, the immune system status, and genetic predisposition are crucial. IBD is most often diagnosed in late adolescence or early adulthood. Due to the availability of quality treatment, the health status of IBD patients also improves, allowing them to consider starting a family after the disease has calmed down. Patients have fewer children than the general population, but there was no significantly higher risk of infertility. A prerequisite for successful pregnancy and childbirth is a stable condition of the disease and the use of treatment during pregnancy to prevent relapse. A specific problem of patients is voluntary childlessness because of worries about the worsening of IBD during or after pregnancy, about negative effects of treatment on a foetus, but also an occurrence of disease in offspring This work aims to summarize the issue of pregnancy in patients with IBD and to provide information on the current situation in patients with IBD in the Czech Republic. Meiosis and fertility of juvenile mouse males

This work is a summary of literature on pecularities of spermatogenesis in juvenile mouse males (Mus musculus) and their utilization. Spermatogenesis is a process that leads through meiosis to sperm production. The cell undergoes in waves the following cell types: spermatogonia, spermatocytes, spermatids, and spermatozoa. Juvenile mice (whose testes size and sperm count have not reached their maximum) are often used to study individual cell types. The transition between cell types takes shorter time in juveniles. Spermatozoa from the 1st wave of spermatogenesis (WS) are derived from prenatal gonocytes, allowing earlier sperm production. They have a lower frequency of crossing over (CO rate) due to a different processing of CO intermediates; the consequence can be aneuploidy (one chromosome less/more). Spermatozoa from the 2nd WS still display lower CO rate. In 3rd WS testes descend and their temperature decreases to 33řC; CO rate is more like in adults. In 4th WS is typical testicular supportive cells mature and CO rate is similar to adult levels. Juvenile males also suffer from more frequent and severe sperm malformations.
